DYNAMICS OF THE MASS COEFFICIENT OF THE LIVER AND KIDNEYS IN SUBCHRONIC CADMIUM INTOXICATION

Abstract

The integral indicator of the general condition of animals during intoxication is the mass coefficient of the organ. The liver and kidneys are the target organs of cadmium. The dynamics of the mass coefficient of the liver and kidneys of experimental sexually mature male Wistar rats with subchronic cadmium intoxication was studied. For 28 days, experimental animals received cadmium acetate with drinking water (15 ml / day) at a dosage of 1/50 LD50. A large variability of this indicator was revealed for the liver, which may be due to its more pronounced participation in the detoxification of cadmium. A statistically significant decrease in the liver mass coefficient of experimental animals compared with the control group was revealed on the 7th, 14th and 21st days of cadmium intake with drinking water, which may be due to inhibition of cell proliferation and synthetic activity. With further intake of cadmium, there are no differences with the control group, which may be due to the activation of various detoxification mechanisms, in particular, the synthesis of metallothioneins in hepatocytes. The relative weight of the kidneys in relation to the body weight of rats when receiving cadmium with drinking water does not change in comparison with control animals.

Relevance. Cadmium is one of the ecotoxicants that leads to a slowdown in the growth and development of organisms, deviation of biochemical parameters, dystrophy and hypertrophy of organs. The liver and kidneys as the main organs of detoxification of xenobiotics are the most sensitive to the action of cadmium [1, 2].
The purpose of the study. To evaluate the effect of subchronic cadmium intoxication of experimental animals on the mass coefficients of the liver and kidneys.
Materials and methods. The experiment was carried out on 50 sexually mature male Wistar rats. To simulate subchronic intoxication, cadmium was administered with drinking water (15 ml/day) in the form of cadmium acetate at a dosage of 1/50 LD50 for 7, 14, 21, 28 days (groups 1, 2, 3, 4). The fifth group in the period 28-56 days and the control group received clean drinking water. The animals were removed from the experiment by instantaneous decapitation, the internal organs were weighed, calculating the mass coefficients of the organs as a percentage of the mass of the organ to the body weight. Statistical processing of the results was carried out using the MS Exel analysis package (p<0.05).
Results. The intake of cadmium acetate leads to a decrease in liver weight relative to the body weight of the animal in groups 1-3 compared with the control group (on the 21st day of intoxication - 3.53% and 3.86%). On the 28th day of intoxication, there were no differences in the relative weight of the liver of experimental and control animals. The administration of cadmium acetate for 28 days did not cause a change in the relative weight of the kidneys in the rats of the experimental groups compared with the control animals.
Conclusions. Subchronic intoxication of animals with cadmium for 21 days leads to a decrease in the relative weight of the liver. With a longer intake of cadmium into the body, a decrease in the mass coefficient of the liver is not observed. At the initial stage of intoxication, there is probably an inhibition of the proliferation of liver cells, in the future there is a restructuring of the metabolism of hepatocytes. Changes in the relative weight of the kidneys with the intake of cadmium acetate in the amount of 1/50 LD50 were not detected.
